Acute hypoxia changes the gene expression profiles and alternative splicing landscape in gills of spotted sea bass (Lateolabrax maculatus)

Hypoxia is one of the most important environmental stressors in aquatic ecosystems. To deal with hypoxia environment, fishes exhibit a series physiological and molecular responses to maintain homeostasis organism functions. In present study, hypoxia-induced changes gene expression profiles alternative splicing (AS) events spotted sea bass ( Lateolabrax maculatus ), promising marine-culture fish species China, were thoroughly investigated by RNA-Seq analysis. A total 1,242, 1,487 1,762 differentially expressed genes (DEGs) identified at 3 h, 6 h 12 gills after stress. Functional enrichment analysis KEGG GSEA demonstrated that HIF signal network system was significantly activated cell cycle process remarkably suppressed response hypoxia. According temporal profiles, six clusters generated protein-protein interaction (PPI) networks constructed for two enriched (cluster 2) or -suppressed 5), respectively. Results showed signaling related including vegfa , igf1 edn1 cox2b cxcr4b ctnnb1 slc2a1a recognized as hubs cluster 2, while mcm2 chek1 pole mcm5 pola1 rfc4 tightly cycle, down-regulated considered 5. Furthermore, 410 differential (DAS) hypoxia, which closely associated spliceosome. Of them, 63 DAS also levels suggesting their might be regulated AS mechanism. This study revealed key biological pathways affected would help us better understand mechanisms other species.
